ObjectiveTo establish a method for simultaneous determination of theophylline and doxofylline in human plasma by RP-HPLC.MethodsPhenomenex C18(250 mm ×4.6 mm,5 μm)column was used.The mobile phase was composed of methanol-water(30∶70)at flow rate of 1.0 ml/min.The detection wavelength was 273 nm and the internal standard was caffeine.ResultsThe linear range was 1.05 to 67.33 μg/ml for theophylline and doxofylline (r>0.999).The recovery rates was 96.79%-105.40%and 91.85%-95.51%respectively.The intra-day precisions and the inter-day precisions were both less than 10%.ConclusionThe method is rapid and accurate.It can be used for the simultaneous determination of theophylline and doxofylline concentration in human plasma.
ObjectiveTo investigate the influence of ozone major autohemotherapy(O-MAHT)on untoward effect related to ischemia reperfusion after angioplasty of lower extremity.Methods75 cases of LEAD patients who had successful endovascular therapy in our hospital from August 2015 to May 2017 were selected as research subjects and divided into 3 groups,25 cases in each group.The patients were treated with O-MAHT three days after angioplasty were included into group A were patients were treated with O-MAHT 12 hours after angioplasty were included into group B;and those had no O-MAHT therapy before and after angioplasty were included into group C.The untoward effect of patients in these three groups after angioplasty was observed and recorded.ResultsThe occurrence rate of untoward effect of patients in group A,group B and group C was 60.0%,64.0%and 88.0%respectively.The group comparison between group A and B regarding the occurrence rate of untoward effect had no statistical difference(P>0.05);and that between group A and C,group B and C had statistical difference (P<0.05).ConclusionApplying O-MAHT to patients who had angioplasty of lower extremity atheroselerotic disease before angioplasty and at the early period of postoperation could protect the lower extremity and reduce the untoward effect due to ischemia reperfusion.
ObjectiveTo discuss the protective effect of Buzhong Yiqi Decoction on liver mitochondrial function in mice infected by candida and its mechanism.Methods40 male Kunming mice were given intraperitoneal injection of candida albicans suspension after pretreatment by CTX and then made into models infected by candida albicans and were randomly divided into the control group(n=20)and experimental group(n=20).Mice in the control group were given Antifungal therapy,while mice in the experimental group were given antifungal drug combined with Buzhong Yiqi Decoction.The survival rate,mitochondriopoiesis index,nitrosative stress index and hepatic pathology were observed.ResultsSurvival rate was significantly higher in the experimental group than that in the control group(70%vs.30%,P=0.011).The level of PGC-1α mRNA was significantly higher in the experimental group than that in the control group(P<0.05).The nitrosative stress index was significantly lower in the experimental group than in the control group,the difference was statistically significant(P<0.05).Conclusionof hepatic sinusoid observed by light microscope was lighter in the experimental group than that in the control group.Conclusion Buzhong Yiqi Decoction may protect the liver mitochondrial function and facilitate mitochondriopoiesis in mice infected by candida albicans via inhibition of nitrosative stress.
