Abstract:Objective To investigate the clinical value of real-time PCR in diagnosis of Mycobacterium tuberculosis(TB) DNA in sputum of patients with pulmonary tuberculosis.Methods Twenty-five patients with tuberculosis or old tuberculosis were enrolled from July to November 2017.Twenty-five healthy subjects were selected as subjects during the same period.Real-time PCR, the acid-fast staining method and the culture method were used to detect the TB of the examinee, and the positive rate of detection and patient satisfaction were observed and evaluated.Results The positive rate in detection of tuberculosis or obsolete tuberculosis of real-time PCR was higher than that of acid-fast staining and culture method, and the difference was statistically significant (P<0.05).The sensitivity of real-time PCR was higher than that of acid-fast staining and culture method, and the difference was statistically significant (P<0.05).There was no significant difference among the quantitative PCR, acid-fast staining and culture method in the specificity (P>0.05).The total satisfaction of the real-time PCR method was higher than that of the acid-fast staining method and the culture method, and the difference was statistically significant (P<0.05).Conclusion Real-time PCR, acid-fast staining and culture methods have high clinical value in the diagnosis of tuberculosis patients, but the quantitative PCR method has the advantages of greater operation, simple and fast operation, and can provide early diagnosis and prevention of diseases, which is worthy of application and promotion.
