基于网络药理学和分子对接研究丹芎活血片治疗骨性关节炎的机制

摘要目的基于网络药理学探讨丹芎活血片治疗骨性关节炎的机制。方法采用数据库及文献筛选丹芎活血片的化合物和靶点、骨性关节炎的基因，映射共同靶点。以Cytoscape 3.7.2 构建蛋白质-蛋白质交互（PPI）网络，在DAVID V6.8 数据库中进行基因本体（GO）和京都基因与基因组百科全书途径富集（KEGG）分析，构建 “成分-靶点-通路” 网络，对关键成分进行分子对接。结果获得成分50 个和疾病基因774 个，GO 功能331 个和KEGG 通路61 条。分子对接结果显示，有效成分为阿魏酸、芥子酸、山柰酚、姜黄素等，关键靶点为STAT3、血管内皮生长因子A（VEGFA）、白细胞介素-6（IL-6）、肿瘤坏死因子（TNF）和JUN 等，KEGG 涉及癌症、乙型肝炎、核因子κB（NF-κB）、PI3K-Akt 和JAK-STAT 等信号通路。结论丹芎活血片可能通过VEGFA、JUN 的表达和PI3K/Akt 信号通路治疗骨性关节炎。

Abstract：Objective To explore the mechanism of Danxiong Huoxue Tablets in the treatment of osteoarthritis based on network pharmacology.Methods The compound and target of Danxiong Huoxue Tablets and the gene of osteoarthritis were screened by database and literature, and the common target was mapped.The protein-protein interaction (PPI)network was constructed using Cytoscape 3.7.2, gene ontology (GO) and Kyoto encyclopedia of genes and genomes pathway enrichment (KEGG) were analyzed in DAVID V6.8 database, and the "component-target-pathway" network was constructed for molecular docking of key components.Results Obtained 50 components and 774 disease genes,331 GO functions and 61 KEGG pathways.Molecular docking results showed that the active ingredients were ferulic acid, sinapic acid, kaempferol, curcumin, etc.And the key targets were STAT3, vascular endothelial growth factor A(VEGFA), interleukin-6 (IL-6), tumor necrosis factor (TNF) and JUN, etc.KEGG was involved cancer, hepatitis B,NF-κB, PI3K-Akt and JAK-STAT signaling pathways.Conclusion Danxiong Huoxue Tablets may treat osteoarthritis through the expression of VEGFA, JUN and PI3K/Akt signaling pathway.
