Analysis of high risk subtypes and related protein expression of human papillomavirus in different types of cervical cancer

Abstract:Objective To analyze the distribution and expression characteristics of high risk human papillomavirus(HPV)subtypes in cervical adenocarcinoma and squamous cell carcinoma.Methods A total of 120 patients with cervical cancer complicated with high-risk HPV infection diagnosed in Jiangmen Central Hospital from January 2018 to February 2020 were selected as the research subjects.HPV subtypes were analyzed by hybrid captune (HC2)method,and the expression of HPV E6,E7,P53 and Rb proteins were detected by immunohistochemical method,the expression differences of HPV high-risk subtypes and E6,E7,P53,Rb protein in cervical adenocarcinoma and squamous cell carcinoma were analyzed,and the correlation between HPV high-risk subtypes and E6,E7,P53,Rb protein expression were analyzed.Results Among the 120 cases of cervical cancer,34 cases were adenocarcinoma and 86 cases were squamous cell carcinoma.The proportion of HPV 18 subtype in cervical adenocarcinoma was higher than that in squamous carcinoma,and the proportion of HPV 16 subtype was lower than that in squamous carcinoma,and the multiple infection rate in cervical adenocarcinoma was higher than that in squamous carcinoma,the differences were statistically significant(P<0.05).There was no significant difference in E6 protein expression between cervical adenocarcinoma and squamous cell carcinoma (P>0.05).The expression of E7 protein in cervical adenocarcinoma was higher than that in cervical squamous cell carcinoma,while the expression of P53 and Rb protein was lower than that in cervical squamous cell carcinoma,the differences were statistically significant(P<0.05).HPV 16 subtype,HPV 18 subtype and positive expression of E6,E7,P53 and Rb protein in cervical cancer patients were positively correlated (P<0.05).Conclusion HPV 16 is the main high-risk HPV subtype of cervical squamous cell carcinoma,and HPV 18 is the main highrisk HPV subtype of cervical adenocarcinoma.There are large differences in the positive expression of E7,P53 and Rb protein between cervical squamous cell carcinoma and adenocarcinoma.Cervical adenocarcinoma Higher multiple infection rate and higher co-expression rate of E6 and E7 may be important factors affecting poor prognosis,and it is worthy of further study and discussion.
