Abstract:Objective To detect the dynamic expression levels of miR-146a and circulating T follicular helper(cTfh)cell during immune response of hepatitis B vaccination,and then analyze the association between miR-146a,cTfh cell and the immune response of hepatitis B vaccination.Methods In July 2017,a group of healthy adults who had completed the standard procedure for the hepatitis B vaccination in Community Health Service Center of Dalang Country were recruited to receive one dose of hepatitis B vaccination,and peripheral blood samples were collected from 12 volunteers at different time points after vaccination,ELISA method was used to measure the concentration of Hepatitis B surface antibody(HBsAb)in serum;Flow cytometry was used to detect the proportion of Tfh cells,B cells,plasma cells,and memory B cells in peripheral blood mononuclear cells(PBMCs).Real-time fluorescence quantitative PCR was used to detect the relative expression of miR-146a.Results cTfh cell reached the highest level(P=0.001)on the 7thday after vaccination,and the HBsAb reached the highest level on the 14thday after vaccination,and the difference was statistically significant(P<0.001).There was a negative correlation between the Tfh cells and B cells(r=-0.426,P=0.03),and a positive correlation between the plasma cells and HBsAb (r=0.330,P=0.04).There was no significant correlation between the expression level of miR-146a,cTfh cell and HBsAb (P>0.05).Conclusion Tfh cells might play a role in the immune response to hepatitis B vaccination,but not demonstrate the association between miR-146a and the immune response of hepatitis B vaccination.
