Influence of strontium ranelate on tibial bone defect healing in rats w ith osteoporosis

Abstract:Objective To discuss the influence of strontium ranelate on tibial bone defect healing in rats with osteoporosis.Methods 30 female SD rats of 3months old were selected and randomly divided into sham operation group(n= 5)and the other 15 rates were given bilateral ovariectomy.At 12 weeks after operation,5 rats were random ly selected as model group.Rats in the sham operation group and the model group were sacrificed,and bone mineral density(BMD)and bonemineral content(BMC)of tibia andmandibular alveolar process were determined.The other 10 ratsweremade bilateral tibial bone defect and randomly divided into two groups.Rats in group A were given strontium ranelate gavage,and those in group B were given same volume of saline gavage.After 12 weeks of intervention,the rats in both groups were sacrificed to obtain their bilateral tibia and mandible model.The healing condition of tibial bone defect was observed,the osteogenesis condition of tibial bone defectwas detected by tissue sections,and BMD and BMC ofmandibular alveolar process were determined.Resu lts At 12 weeks of operation,the weight gain was significantly higher in the model group than that in the sham operation group(P<0.01),and the BMD and BMC were significantly lower in the model group than those in the sham operation group(P<0.01).At 12 weeks of drug intervention after tibial bone defect operation,general healing of tibia could be observed.The general observations between group A and group B showed no significant difference.Results of HE staining showed that osteogenesis wasmore significant at bone defect in group A than that in group B,and BMD and BMC of mandibular alveolar process were significantly higher in group A than in group B(P<0.01).Conclusion Strontium ranelate has a positive effect on bone defecthealing in ratswith osteoporosis.
