Effect of Total Flavonoids of Rhizoma Drynariae on BMP-2 protein expression and serum bone gla protein level in rats with osteoporosis

Abstract:Objective To explore the effect of Total Flavonoids of RhizomaDrynariae on BMP-2 protein expression and serum bone gla protein level in rats with osteoporosis.Methods Osteoporosis model was established by removing rat ovary,and the rats were divided into normal group,sham operation group,model group,total Flavonoids of Rhizoma Drynariae group and Caltrate D Tablet group.In the seventh days after surgery,drugs were used once per day for 12 consecutive weeks.Morphological change of bone tissue in rats was observed by HE staining,and the expression of BMP-2 protein and the level of serum bone gla protein level in the femur of rats in each group was detected by ELISA.Results HE staining indicated that the number,density,distribution,and integrity of bone trabecula in the rats femoral bone tissue in the Total Flavonoids of Rhizoma Drynariae group were superior to those in the model group.The protein expression of BMP-2 in rats femoral bone tissue and the level of serum bone gla protein [(13.469±0.215)pg/ml and (1.61± 0.09)μg/L respectively]was much higher than that in the model group[(5.516±0.119)pg/ml and(0.76±0.22)μg/L respectively](P<0.05).The protein expression and serum bone gla protein level was close to that in the sham operation group respectively[(15.279±0.211)pg/ml and(1.34±0.17)μg/L](P>0.05).Conclusion In the treatment of rats with osteoporosis,Total Flavonoids of Rhizoma Drynaria can obtain a great effect.The possible mechanism is through up-regulation of BMP-2 protein in the femoral bone tissue and the level of serum bone gla protein,thus promoting the formation of bone tissue.
