Abstract:Objective To observe the effect of nerve growth factor(NGF)and its low affinity receptor(p75 neurotrophin receptor,p75NTR)on the apoptosis and proliferation of rat hepatic stellate cells (HSC),and to explore the possible mechanism of apoptosis.Methods Rat hepatic stellate cells(HSC-T6)were incubated in vitro,HSC-T6 was incubated with NGF and NGF+p75NTR with different concentrations.The cells were divided into the control group,the NGF group and the NGF+p75NTR group.The control group was cultured with 0 ng/ml NGF+0 ng/ml p75NTR,the NGF group was cultured with 100 ng/ml NGF,and the NGF+p75NTR group was cultured with 100 ng/ml NGF+100 ng/ml p75NTR.The flow cytometry was used to detect the apoptosis condition.The effect of HSC on the proliferation of NGF and p75NTR was observed by XTT,the expression of P53 and Caspase-3 were detected by immunohistochemical staining.Results The inhibitory effect of the NGF group on the proliferation of HSC was statistically significant(P<0.05)with the NGF+p75NTR group and the control group (P<0.05).The flow cytometry results showed that the apoptosis rate of the control group was lower than that of the NGF group and the NGF+p75NTR group(P<0.05),the apoptosis rate of the NGF+p75NTR group was higher than the NGF group(P<0.05).The expression of P53 and Caspase-3 in the NGF group and the NGF+p75NTR group were significantly higher than that in the control group (P<0.05),the expressions in the NGF+p75NTR group were significantly higher than that in the NGF group(P<0.05).Conclusion NGF may inhibit HSC-T6 proliferation and promote its apoptosis through p75NTR,which may be related to the regulation of P53 and Caspase-3 expression.
