Effect of acupuncture on neuronal apoptosis and adenosine level in rats with transient ischemic attack

Abstract:Objective To study the effect of acupuncture on apoptosis and adenosine level of neurons in rats with transient ischemic attack(TIA).Methods 32 male Wistar rats are randomly divided into 4 groups,normal control group(8 rats),sham-operation group(8 rats),model group(8 rats),acupuncture group(8 rats).Global cerebral ischemia TIA rat model was established by four-vessel occlusion.The acupuncture group was adminstered with electro acupuncture therapy and other 3 groups without any other treatment,to sample 1 week after surgery.By the TUNEL dyeing test(In situ terminal labeling)to detect the Neuron apoptosis in cerebral cortex and hippocampus CA1 area,and determine Adenosine content in cerebral cortex and hippocampus area by spectrophotometer.Results The neuronal apoptosis in the CA1 area and cerebral cortex of the model group were (36.44±8.96),(36.94±8.77)single/mm2higher than that of the acupuncture group (18.52±6.32),(20.11±6.70)single/mm2 with significant difference(P<0.05).As for normal control group and the sham operation group,the values were(13.98±5.47),(11.63±4.55)single/mm2 both lower than that of model group,with statistical difference (P<0.05).Adenosine content: the CA1 area and cerebral cortex of the model group were(0.139±0.016)and(0.140±0.017)μmol/L mg,lower than that of the acupuncture group(0.176±0.004)(0.178±0.006)μmol/L mg,with statistical difference(P<0.05).As for normal control group(0.038±0.006),(0.039±0.009)μmol/L mg and the sham operation group(0.037±0.006),(0.038±0.005)μmol/L mg were lower than the model group,and the difference was statistically significant(P<0.05).Conclusion Acupuncture can inhibit the apoptosis of neurons in TIA rats and increase the content of endogenous adenosine in them.
