Application of loop-mediated isothermal amplification for the detection of mycobacterium tuberculosis

Abstract:Objective To establish the loop-mediated isothermal amplification technique (LAMP)for the detection of Mycobacterium tuberculosis(MTB)gene in the sputum of patients with pulmonary tuberculosis.Methods Three pairs of specific primers were designed based on highly conserved 16S rDNA of MTB as the target gene.From January 2017 to December 2017,89 sputum samples from the patients were with clinically diagnosed pulmonary tuberculosis,20 sputum samples from non-tuberculosis patients and the MTB standard strain(H37RV)were collected.LAMP method was established,and its specificity and sensitivity were tested.The MTB culture results were taken as the gold standard.SPSS13.0 was used for data processing and statistical analysis.Results LAMP was successfully established for the detection of MTB in the sputum.Its specificity was 100%and its sensitivity was 50 copies.In the clinical sputum testing,the specificity and sensitivity were 85%and 96.63%respectively.The results were almost identical with those of the MTB culture,the difference was statistically significant(P>0.05).Conclusion LAMP has high specificity and high sensitivity in the detection of MTB,which is suitable for the rapid detection of MTB in primary medical institutions.
