Abstract:Objective To investigate the protective effects of application of Angiotensin-1(Ang-1)on lung injury and inflammatory responses induced by lipopolysaccharide(LPS).Methods Altogether 30male KM mice were randomly divided into three groups(control group,LPSgroup and Ang-1 group),with 10mice in each group.Intratracheal instillation of LPSwas performed so as to constructan acute lung injury(ALI)model.Either recombinant adenovirus-green fluorescent protein(LPSgroup)or recombinant adenovirus-human angiotensin 1 was intravenously injected intomice by vena caudalis 24 h beforemodeling;control group was given normal saline.All themice were sacrificed 24 h after the establishment of ALI so as to determine the Wet-Dry(W/D)ratio of lung,the histological changes of lung tissues was observed and serum interleukin-6(IL-6)level and granulocytemacrophage-colony stimulating factor(GM-CSF)level in bronchoalveolar lavage fluid(BALF)according to ELISA were detected.Results TheW/D ratio of lung and serum IL-6 level of Ang-1 group were(4.05±0.16)and(173.07±10.05)ng/ml,significantly lower than(5.46±0.07)and(324.61± 8.92)ng/ml in LPS group;the GM-CSF level in BALF of Ang-1 group was(15.05±1.21)ng/ml,significantly higher than that in LPSgroup(10.24±1.35)ng/ml.At the same time,the pathological injury of lung tissues in Ang-1 group was significantly reduced compared with LPS group.Conclusion Ang-1 can reduce the lung tissue damage and inflammatory responses induced by LPS.
