Study on the effect mechanism of scutellarin in the occurrence and development of colon cancer

Abstract:Objective To explore the effect mechanism of scutellarin in the occurrence and development of colon cancer.Methods MTT, flow cytometry, wound healing, Transwell chamber and mouse tumor-bearing test were carried out to determine the effect of scutellarin on the proliferation, apoptosis, migration, invasion and tumorigenesis of colon cancer T84 cells respectively.Western Blot was performed to assess the protein levels of Bcl-2/Bax, caspase3/9, ERK1/2,p-ERK1/2, epidermal growth factor receptor (EGFR), c-myc and c-fos after T84 cells were treated with scutellarin.Immunofluorescence technique was used to evaluate the subcellular location of ERK1/2 in T84 cells after scutellarin treatment.Results Scutellarin treatment inhibited the proliferation, migration, invasion and tumorigenesis of T84 cells,and induced cell apoptosis;scutellarin treatment decreased the expression of p-ERK1/2, EGFR, c-myc and c-fos, while translocated ERK1/2 from nuclear to cytoplasm.Inhibition of cell proliferation and tumorigenesis and enhancement of cell apoptosis caused by scutellarin were all weakened when EGFR was up-regulated in T84 cells.All differences were statistically significant (P<0.05).Conclusion Scutellarin represses the progression of colon cancer through inhibiting the activation of EGFR/ERK1/2 signaling pathway.
