Abstract:Objective To study the expression changes of axon guidance factor Netrin-1 and its effect on the migration and apoptosis of hepatocellular carcinoma cells HepG2 under hypoxia.Methods HepG2 cells were cultured in 1% O2 anoxic environment.The expression of Netrin-1 in HepG2 cells under hypoxia was detected by Western-blot.Interfering plasmid shRNA-Netrin-1 mRNA was constructed and stably transfected into HepG2 cells.Transwell method and flow cytometry were used to observe the changes of cell migration and invasion and apoptosis rate of HepG2 cells under normal and hypoxia conditions.Results The expression level of Netrin-1 protein in HepG2 cells was increased after 24 h hypoxia culture,cell migration experiments indicated hypoxia promoted the migration of HepG2 cells,flow cytometry results showed that the apoptosis of HepG2 cells was inhibited by hypoxia,the differences were statistically significant(P<0.05).The Netrin-1 knockdown significantly inhibited the hypoxia induced HepG2 cell migration,the number of permeable cells in shRNA-Netrin-1-HepG2 cells decreased,and the Netrin-1 knockdown could also increase HepG2 cell apoptosis ratio,the differences were statistically significant(P<0.05).Conclusion Hypoxia can promote hepatocellular carcinoma cell Netrin-1 expression,and hypoxia promotes the migration of hepatocellular carcinoma cells HepG2 and inhibits their apoptosis,which may be related to the increased expression of Netrin-1.
