Effect of NGF-BMSCs transplantation on the improvement of symptoms in AD animal model and the levels of apoptosis-related genes Bcl-2and Bax in brain tissue

Abstract:Objective To investigate the effect of nerve growth factor-bone marrow stromal stem cells (NGF-BMSCs)transplantation on the improvement of symptoms in (AD) animal model and the levels of apoptosis-related genes B lymphocyte tumor-2 (Bcl-2) and Bax in brain tissue.Methods Fifty AD rats were randomly divided into five groups,namely the control group, saline injection group, model group, BMSCs transplantation group and NGF-BMSCs transplantation group, with 10 rats in each group.The behavioral changes of AD rats were observed, and the levels of Bcl-2and Bax in hippocampus of rats were detected.Results The escape latency of the model group and the saline injection group from the first day to the sixth day was shorter than that of the control group, with statistically significant differences (P<0.05).The escape latency of BMSCs transplantation group and NGF-BMSCs transplantation group from the first day to the sixth day was longer than that of the control group, model group and saline injection group, with statistically significant differences (P<0.05).The escape latency of the NGF-BMSCs transplantation group from the first day to the sixth day was longer than that of the BMSCs transplantation group, and the difference was statistically significant(P<0.05).The levels of Bcl-2and Bax in the model group, saline injection group, BMSCs transplantation group, and NGF-BMSCs transplantation group were higher than those in the control group, and the differences were statistically significant (P<0.05).The Bcl-2levels in the saline injection group and the BMSCs transplantation group were higher than those in the model group, the Bax levels were lower than those in the model group, and the differences were statistically significant (P<0.05).The levels of Bcl-2in the BMSCs transplantation group and NGF-BMSCs transplantation group were lower than those in the saline injection group, and the differences were statistically significant (P<0.05).The Bcl-2level in the NGF-BMSCs transplantation group was lower than that in the BMSCs transplantation group, the Bax level was higher than that in the saline injection group and in the BMSCs transplantation group, and the differences were statistically significant (P<0.05).Conclusion NGF-BMSCs transplantation can inhibit neuronal apoptosis in AD animal models by regulating the expression levels of Bcl-2and Bax in hippocampus.
