Abstract：Objective To explore the influence of Irbersartan on HGF level in myocardial infarction rats.Methods Myocardial infarction (MI)model was built by ligating the left anterior descending coronary.The survived rats after 24 hours were randomized into the Irbersartan group (n=11)and the control group (n=9),in addition,the sham operation group(n=6)was set up.The Irbesartan group was given Irbesartan(50 mg/[kg·d]).The control group and the sham operated group received the same volume physiological saline by intraperitoneal injection for 4 weeks,the blood was harvested from the posterior orbital vein for 24 hours,and 1,2,4 weeks after surgery,the level of HGF were determined by ELISA.Rats were sacrificed after 4 weeks,and HE staining was used to observe the pathological changes of ischemic myocardium.Results HE staining revealed that the myocardial cells were arranged neatly in the sham operation group,a small amount of inflammatory cell infiltrated.Myocardial cells were dissolving and myocardial structure disordered in the Irbersartan group and the control group,the pathological changes in the Irbersartan group were less than those in the control group.The level of HGF at 24 hours after operation was detected in all groups rats,and there was no significant difference between the each groups(P＞0.05).The level of HGF in the sham operated group after 1 week was lower than that at 24 hours after operation(P＜0.01),and which went up at 2 and 4 weeks,but which was still lower than that at 24 hours after operation (P＜0.01).The levels of HGF in the control group and the Irbesartan group at 1 week were higher than those at 24 hours,and the level of HGF in the control group and the Irbesartan group decreased significantly at 4 weeks,which was lower than that at 1 weeks(P＜0.01).At 1 and 2 weeks,the levels of HGF in the control group and the Irbesartan group were higher than those in the sham operation group (P＜0.01),at 2 and 4 weeks,the level of HGF in the Irbesartan group was significantly lower than that in the control group (P＜0.01).The level of HGF in the control group at 1 and 2 weeks was higher than that at 24 h after operation(P＜0.01),the level of HGF in the control group at 2 and 4 weeks was higher than that at 1 weeks(P＜0.05,P＜0.01).The level of HGF in the Irbesartan group at 4 weeks was lower than that at 24 h and 1 weeks after operation (P＜0.05,P＜0.01).Conclusion Irbersartan can improve the pathological changes of ischemic myocardium for 4 weeks.The serum level of HGF is increased in MI rats,and peaked at 1 week.Irbesartan can reduce serum HGF level after 4 weeks treatment.
