Abstract:Objective To compare the differences and advantages between different methods of chronic myocardial ischemia.Methods Thirty male ICR mice were selected and divided into subcutaneous injection group,sustained-release pump release group and control group according to different modeling methods,with 10 rats in each group.In the subcutaneous group,Isoproterenol (ISO) 30 mg/(kg·d) was injected subcutaneously into the lumbar back of the mice for 14 days.The release pump group was placed subcutaneously in the lumbar back of the mice.The ISO release amount was 30 mg/(kg·d).The control group was injected subcutaneously with an equal dose of physiological saline.Myocardial ischemia was measured by myocardial zymogram and heart section TTC staining.Results There was no significant difference in the mortality between the subcutaneous injection group and the sustained-release pump release group (P>0.05).The cardiac hypertrophy index of the mice in the subcutaneous injection group and the sustained-release pump release group was higher than that in the normal control group,and the difference was statistically significant (P<0.05).There was no significant difference in the cardiac hypertrophy index between the subcutaneous injection group and the sustained-release pump release group (P>0.05).The myocardial ischemic area of the mice in the sustained-release pump release group was higher than that of the subcutaneous injection group,and the difference was statistically significant(P<0.05).The myocardial enzymes and cardiac troponin T (cTnT) level of the mice in the sustained-release pump release group were higher than those in the normal control group,and the differences were statistically significant (P<0.05).The levels of α-hydroxybutyrate deoxygenase (α-HBDH),aspartate aminotransferase (AST),isozyme (mAST) and cTnT in the subcutaneous group were higher than those in the normal control group,and the differences were statistically significant (P<0.05).While the levels of creatine kinase (CK) and isoenzyme (CK-MB) and lactate deoxygenase(LDH) in the subcutaneous group were not significantly different from those of the normal control group (P>0.05).There was no significant difference in myocardial zymogram and cTnT between the sustained-release pump release group and the subcutaneous injection group (P>0.05).Conclusion Both subcutaneous injection and sustained release pump release ISO can cause myocardial hypertrophy in mice,sustained release pump can induce myocardial ischemia in mice to a greater extent,and sustained release pump also causes damage to myocardial function to a greater extent.
