Abstract:Objective To explore the injury mechanism of Toll-like receptor 3 (TLR3) gene rs3775291 mutation in acute and chronic myocardial ischemia of diabetes mellitus rats.Methods Eighty gene knockout SD rats were selected as the research objects from the Experimental Animal Center of Nanchang University from January to April 2019.They were divided into wild-type group (n=40) and mutant type group (n=40) by random number table method.The rats in the wild-type group were only silk thread passed through coronary artery but left ventricular branch was not ligated,and normal saline was injected through tail vein.In the mutant type group,TLR3 gene knockout rats were selected to make the model.The TLR3,nuclear factor κB (NF-κB) protein expressions,chemokine 10 (IP-10),interleukin 8 (IL-8),and tumor necrosis factor α (TNF-α) levels were compared between the two groups.Results The TLR3 and NF-κB protein expressions in the mutant type group were higher than those in the wild-type group,with statistically significant differences (P<0.05).There were no significant differences in the levels of IP-10,IL-8 and TNF-α between the two groups (P>0.05).Conclusion In the diabetes mellitus rat model of acute and chronic myocardial ischemia,the expression levels of TLR3 gene and protein were significantly increased,and the levels of inflammatory indicators were also increased.The mutation of TLR3 gene rs3775291 site is closely related to the injury mechanism.
