Effect of Duhuojisheng Decoction on immune function of collagen induced arthritis rats

Abstract:Objective To explore the effect of Duhuojisheng Decoction on the immune function of collagen induced arthritis (CIA) rats.Methods A total of 50 clean male Wistar rats were fed adaptively for 7 days, six rats were randomly selected as the normal group, the other rats were injected with 200 μl collagen emulsion into the skin of the left plantar part of their feet to make CIA rat model.Arthritis index(AI)≥4 was used as the model successfully.The model rats were randomly divided into model group,Celecoxib(0.05 g/[kg·d])group,Duhuojisheng Decoction high concentration (44 g/[kg·d])group, Duhuojisheng Decoction medium concentration (22 g/[kg·d]) group and Duhuojisheng Decoction low concentration (11 g/[kg·d]) group, The control group and model group were treated with distilled water of equal volume, once a day, and gavage for 30 days.The levels of T cell specific transcription factor (T-bet), interleukin-2 (IL-2), interferon-γ(IFN-γ), GATA binding protein 3 (GATA-3), interleukin-6 (IL-6) and interleukin-13 (IL-13) in serum of six groups of rats were detected and compared by double antibody sandwich enzyme-linked immunosorbent assay (ABC-ELISA).Results The levels of T-bet, IL-2 and IFN-γ in serum of the model group were higher than those of the normal group,and the levels of GATA-3, IL-6 and IL-13 were lower than those of the normal group, the differences were statistically significant (P<0.05).The T-bet level of Celecoxib group and Duhuojisheng Decoction low, medium and high concentration group were lower than those of model group, the level of IL-2 in Celecoxib group and Duhuojisheng Decoction medium group were lower than those in model group, the IFN-γ levels of Celecoxib group, Duhuojisheng Decoction low and medium concentration group were lower than those of model group, the levels of GATA-3, IL-6 and IL-13 in Celecoxib group, Duhuojisheng Decoction low, medium and high concentration group were higher than those in model group, the differences were statistically significant (P<0.05).Conclusion Duhuojisheng Decoction can exert ideal therapeutic effect on CIA rats.Its anti-inflammatory mechanism is to reduce T-bet levels, increase GATA-3 levels, and suppress immune inflammatory factor levels by regulating the Th1 / Th2 cell balance.
