Influence of Berberine on PI3K/Akt/eNOS signal transduction pathway in thoracic aorta of type 2 diabetic rats

Abstract:Objective To observe the influence of Berberine on glucose and lipid metabolism and PI3K/Akt/eNOS signaling pathway of thoracic aorta in diabetic rats,and to investigate the possible protective mechanism of Berberine on macroangiopathy.Methods A total of 60 SD rats were randomly divided into the normal group (n=10)and the experimental group (n=50).The model of type 2 diabetes mellitus was established by intraperitoneal injection of Streptozotocin combined with high-fat and high-sugar diet.The diabetic rats were randomly divided into the model group (n=10),the Metformin group(n=10),and the Berberine group(n=10).The Metformin group and the Berberine group were given Berberine and Metformin respectively,the normal group and the model group were given normal saline by oral gavage.After 8 weeks,fasting blood glucose (FBG),serum concentrations of triglyceride (TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-c),low density lipoprotein cholesterol(LDL-c)and nitric oxide (NO)were measured.The histological morphology of the thoracic aorta was observed by HE staining,and the protein expression of p-PI3K-p85,p-Akt and p-eNOS was detected by Western blotting.Results The levels of FBG,TC,TG and LDL-c in the model group,the Berberine group and the Metformin group were higher than those in the normal group,the levels of HDL-c and NO,the expression of p-PI3K-p85,p-Akt and p-eNOS in thoracic aorta in the model group,the Berberine group and the Metformin group were lower than those in the normal group,with significant difference (P<0.01).The levels of FBG,TC,TG and LDL-c in the Berberine group and the Metformin group were lower than those in the model group,the levels of HDL-c and NO,the expression of p-PI3K-p85,p-Akt and p-eNOS in thoracic aorta in the Berberine group and the Met-formin group were higher than those in the model group,with significant difference(P<0.01).Compared with the model group,the macrovascular endothelium injury in the Berberine group and the Metformin group was relieved.Conclusion The protective effect of Berberine on macroangiopathy in diabetic rats may be related to improving glucose and lipid metabolism and activating PI3K/Akt/eNOS signaling pathway.
