Association analysis of BMI-1 and MMP-3 gene polymorphisms with breast cancer susceptibility in Han nationality

Objective To study the association between three single nucleotide polymorphisms of B-cell-specific moloney leukemia virus insection site 1(BMI-1)and matrix metalloproteinase 3(MMP-3)genes and breast cancer susceptibility in Han nationality.Methods Using the case-control study method,from January 2014 to July 2018,a total of 524 Han women with breast cancer treated in the Second Affiliated Hospital of Qiqihar Medical College,the Third Affiliated Hospital of Qiqihar Medical College,Qiqihar Jianhua Hospital,and Daqing Oilfield General Hospital were selected as the case group,and 518 healthy people receiving healthy checkup in the same period were selected as the control group.PCR-LDR technology was used to detect and type the genetic polymorphisms of BMI-1 gene rs1042059,rs201024480 locus and MMP-3 gene rs3025058 locus,and the risk value(OR)and 95% confidence interval(CI)were used to express the risk of breast cancer.Results There were statistically significant differences in allele frequency and genotype distribution of MMP-3 gene rs3025058 locus between the case group and the control group(P=0.008).There was no statistically significant difference in gene polymorphism of BMI-1 gene rs1042059 locus and rs201024480 locus between the case group and the control group(P>0.05).After adjusting for confounding factors,the risk of breast cancer in the population carrying allele 5A and genotype 5A5A was increased by 1.51 and 1.48 times respectively(OR=1.51,95%CI: 1.17-1.94; P=0.001 and OR=1.48,95%CI: 1.16-1.91; P=0.001).Conclusion The polymorphism of MMP-3 gene rs3025058 is associated with the risk of breast cancer,and can be used as a candidate locus for breast cancer
