Intervention effect of Rhizoma Drynariae Total Flavone on osteoporosis in ovariectomized rats

Abstract:Objective To explore the intervention effect of Rhizoma Drynariae Total Flavone on osteoporosis (OP) in rats.Methods A total of 21 Sprague-Dulle (SD) rats were selected and divided into blank group, model group and treatment group according to the random number table method, with 7 rats in each group.The blank group was left untreated, ovariectomy-induced osteoporosis model was established in the model group, and the treatment group was administered with 0.054 g/(kg·d) Rhizoma Drynariae Total Flavone after model establishment.The bone mineral density(BMD) of the right femur was measured by the dual-energy X-ray absorptiometry.The expression levels of bone transformation markers osteocalcin (BGP), the bone alkaline phosphatase (BALP) and the thyroxine (PTH) in serum were detected by the enzyme-linked immunosorbent assay (ELISA), and the histopathological changes of bone hematoxylineosin (HE) staining were observed by microscope.Results Before the intervention, the BMD of the model group was lower than that of the blank group, with statistically significant difference (P<0.05).There was no significant difference in BMD between the treatment group and the model group before the intervention (P>0.05).After the intervention, the BMD of the blank group and the treatment group was higher than that of the model group, with statistically significant difference (P<0.05).The BMD in the treatment group was lower than that in the blank group after the intervention, and the difference was statistically significant (P<0.05).There was no statistically significant difference in BMD among the three groups before and after the intervention (P>0.05).The HE staining showed that the number, density, layout and integrity of trabeculae in the femoral bone tissue of the treatment group were better than those of the model group.The ELISA results showed that the BGP in the model group was lower than that in the blank group, BALP and PTH were higher than those in the blank group, and the differences were statistically significant (P<0.05).The BGP of the treatment group was lower than that of the blank group, BALP and PTH were higher than those of the blank group, and the differences were statistically significant (P<0.05).The BGP of the treatment group was higher than that of the model group, BALP and PTH were lower than those of the model group, and the differences were statistically significant (P<0.05).Conclusion The Rhizoma Drynariae Total Flavone intervention has a good effect on the OP induced by castration.
