Comparative study on the intervention mechanism of vibration and electroacupuncture on motor dysfunction in young rats with hypoxic-ischemic encephalopathy

Abstract:Objective To compare the effect of whole-body vibration training and electroacupuncture on Bederson score,gastrocnemius muscle fiber morphology and gastrocnemius muscle wet weight ratio in young rats with hypoxic-ischemic encephalopathy (HIE)from the perspective of Bcl-2 anti-myocyte apoptosis regulated by NF-κB,and to study the formation and treatment mechanism of motor dysfunction in young rats with HIE.Methods A total of 120 7-dayold healthy SD young rats were divided into 4 groups by random number table method.No modeling and any intervention were done in sham operation group.HIE model was made in the other three groups:the model control group was bound every day;the electroacupuncture treatment group was treated with electroacupuncture;the vibration treatment group was given vibration treatment for 4 weeks.Bederson scores,fibrous morphology,NF-κB gene and Bcl-2 mrna expression levels of gastrocnemius muscle were compared before modeling,2 weeks after modeling and after treatment.Results After 2 weeks of modeling,Bederson scores of model control group,electroacupuncture group and vibration group were higher than those of sham op-eration group,and the differences were statistically significant(P<0.05).After treatment,the Bederson scores of electroacupuncture treatment group and vibration treatment group were lower than those of model control group,and the differences were statistically significant(P<0.05).After treatment,there was no statistically significant difference in Bederson score between the electroacupuncture treatment group and the vibration treatment group(P>0.05).After treatment,the wet weight of gastrocnemius in vibration treatment group and electroacupuncture treatment group were higher than that in model control group,and the differences were statistically significant(P<0.05).The proportion of wet weight of gastrocnemius muscle in body weight in vibration treatment group was higher than that in electroacupuncture treatment group,and the difference was statistically significant(P<0.05).After treatment,the gastrocnemius muscle fibers in the model control group shrank significantly and the muscle fibers degenerated,the muscle fibers in the vibration treatment group and electroacupuncture treatment group were closely arranged and the muscle transverse lines were regular versus those in the model control group,the nucleus of muscle fibers in the electroacupuncture treatment group increased significantly.After treatment,the positive expression of Bcl-2 mRNA and NF-κB gene in model control group,vibration treatment group and electroacupuncture treatment group were lower than those in sham operation group,and the differences were statistically significant(P<0.05).The positive expression of Bcl-2 mRNA and NF-κB gene in vibration treatment group and electroacupuncture treatment group were higher than those in model control group,and the differences were statistically significant(P<0.05).The positive expression of Bcl-2 mRNA and NF-κB gene in electroacupuncture treatment group were higher than those in vibration treatment group,and the differences were statistically significant(P<0.05).Conclusion Regulation of Bcl-2 expression by NF-κB gene for reduction of the anti-apoptotic ability of skeletal muscle cells is one of the reasons for skeletal muscle fiber injury,muscle atrophy and motor dysfunction in HIE young rats.Compared with vibration therapy,electroacupuncture can better activate NF-κB,regulate Bcl-2 anti-apoptotic expression,improve the anti-apoptotic and multiplication capacity of skeletal muscle cells,repair muscle cell damage,increase the number of muscle fibers,alleviate muscle atrophy and improve the motor dysfunction of HIE young rats.
