Influence of Nonylphenol on ovary and serum estradiol and progesterone level in mice

Abstract:Objective To study the influence of Nonylphenol(NP)poisoning by intragastric administration on ovary,estrogen(E2)and progesterone(P)level in Kunming mice,in order to evaluate the reproductive and developmental toxicity of NP.Methods Thirty healthy SPF female mice of Kunming were randomly divided into the control group(corn oil)and 25,100 mg/kg NP group,there were 10 mice in each group.Gastric administration was used for poisoning,the contamination capacity was 0.1 ml/10 g,once a day,5 days a week,12 weeks in a row.Serum E2,P and FSH were measured and ovarian histopathological sections were observed.Results There was no significant difference in serum E2,P and FSH between the 25 mg/kg NP group and the control group(P>0.05),and the number of primary follicles and mature follicles in ovarian histopathological sections of 25 mg/kg NP group and control group was significantly different(P<0.05).The level of E2,P and FSH in serum and the number of primary follicles and mature follicles in pathological sections of ovary between 100 mg/kg NP group and control group,25 mg/kg NP group were significantly different(P<0.05),and the primary follicles atrophied,the granulosa cells of ovary loosened,the nucleus of oocyte condensed,and the follicular cavity became smaller in 100 mg/kg NP group.Conclusion Increased dose of NP can cause damage to ovarian function in mice having reproductive toxicity and developmental toxicity.
