Abstract:ObjectiveTo investigate the effect of Tangerine Peel,Eucommia and Hawthorn mixture decoction on reducing fat in obese rats induced by high fat diet.MethodsA total of 50 Wistar male rats who fed with high fat diet to establish the rat model of obesity were selected as subjects,which were divided into model group,positive group,low-dose group,middle-dose group and high-dose group randomly,10 rats in each groups.In addition,10 normal feeding rats were selected as the control group.The low-dose group,the middle-dose group and the high-dose group were treated with 1 ml/100 g,2 ml/100 g,4 ml/100 g herbal mixture decoction respectively each day.The rats in the positive group were intragastric Orlistat Suspension 60 mg/kg by gavage.All were treated continuously for 8 weeks.The model group and the control group were given normal saline by gavage with the same method.The content of serum triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C)in serum and the contend of fatty acid synthase(FAS),acyl coenzyme A oxidase(ACO)in liver were detected by enzyme-linked immunosorbent assay.The number of intestinal flora of rats in each group were compared.ResultsThe weight,Lee′s index,TG,TC,LDL-C and FAS contents of the rats in the model group were significantly higher than those in the control group,and HDL-C and ACO of the rats in the model group were lower than those in the control group,the differences were statistically significant(P<0.05).The weight and Lee′s index,TG,TC,LDL-C and FAS contents of the rats in the high-dose group were significantly lower than those in the model group,and the levels of HDL-C and ACO of the rats in the high-dose group were significantly higher than those in the model group,the differences were statistical-ly significant(P<0.05).The number of bifidobacterium,lactobacillus and enterococcus in the intestinal tract of rats in high-dose group were higher than those in the model group,and the number of escherichia coli was less than that of the model group,the differences were statistically significant(P<0.05).There were no statistically significant differences in Lee′s index,TG,TC,LDL-C,FAS and ACO of rats in high-dose group compared with the positive group (P>0.05).ConclusionThe three herbs mixed decoction of Tangerine Peel,Eucommia and Hawthorn can inhibit the activity of FAS,promote the activity of ACO and increase the number of probiotics.It had a significant effect on reducing weight and blood lipids in obese rats induced by high fat diet.
