ComParison of analgesic and anti-Inflammatory effect of Resina Draconis generated by different artificial induction methods

Abstract:Objective To make an investigation on the comparison of analgesic and anti-inflammatory effects of Resina Draconis generated by different artificial induction methods.Methods The experimental data was derived from the laboratory of pharmaceutical factory of Guangxi University of TCM,which was collected in December 2015.The writhing test method was adopted in the analgesic test that recruited and divided 80 mice into 8 groups randomly and evenly with 10 mice in each group,including Saline Group,Aspirin Group (0.5 g/kg),Large-Dosage Natural Group and Small-Dosage Natural Group(naturally-produced Resina Draconis with small dosage and with large dosage,1 g/kg and 2 g/kg),Large-Dosage Biological Group and Small-Dosage Biological Group (biological-induction-produced Resina Draconis with small dosage and with large dosage,1 g/kg and 2 g/kg),Large-Dosage Chemical Group and Small-Dosage Chemical Group(chemicalinduction-produced Resina Draconis with small dosage and with large dosage,1 g/kg and 2 g/kg).The drug was administered orally to mice in each group atthe dose of 0.1 ml/10 g,1 time a day,for 3 consecutive days,the effect of each drug on chemically-stimulated pain reactions(the number of writhing times,writhing latency and the inhibition rate)were observed.As for the anti-inflammatory test,50 mice were recruited and divided into 5 groups randomly and evenly with 10 mice in each group,including Control Group (administered with the same volume of distilled water),Aspirin Positive Control Group (0.2 g/kg),Natural Group(naturally-produced Resina Draconis,2 g/kg),Biological Group(biological-induction-produced Resina Draconis,2 g/kg)and Chemical Group (chemical-induction-produced Resina Draconis,2 g/kg).The drugs were continuously and intragastrically administered to all the mice one time a day,for 7 days.The right ears of all the mice were coated with 20 μl xylene 1 hour after the last administration to make all the mice′s right ears swelling.All the mice were killed 1 hour later and their ears were cut off.Then,the ear flaps were cut off with a 6 mm punch and weighed to calculate the weight gaps between the right ear and the left ear and observe the anti-inflammatory effect(the swelling degree of ears).Results According to the writhing test results,the number of writhing times in Large-Dosage Natural Group and Small-Dosage Natural Group,Large-Dosage Biological Group and Small-Dosage Biological Group,Large-Dosage Chemical Group and Small-Dosage Chemical Group as well as Aspirin Group were(9.01±6.87),(10.05±7.87),(10.51±6.37),(11.05± 7.57),(10.39±7.35),(10.15±7.12),(10.00±4.57)times versus Saline Group′s of(21.10±10.62)times,which showed obvious inhibitory effects on the number of writhing times(P<0.01)in all the groups except Saline Group.Large-Dosage Natural Group,Small-Dosage Natural Group,Large-Dosage Biological Group,Small-Dosage Biological Group,Large-Dosage Chemical Group and Small-Dosage Chemical Group demonstrated similarities with Aspirin Group in the analgesic effects(P>0.05).No significant difference was seen between the writhing frequency of Large-Dosage Natural Group,Small-Dosage Natural Group,Large-Dosage Biological Group,Small-Dosage Biological Group,Large-Dosage Chemical Group and Small-Dosage Chemical Group (P>0.05).As for the anti-inflammatory experiment,the swelling degree of ears in Natural Group,Biological Group,Chemical Group,Aspirin Positive Control Group and Control Group were(7.8±2.3),(7.3± 2.6),(7.5±2.8),(7.0±2.2),(11.2±2.4)mg respectively,with the existence of significant difference between Control Group and the other four groups(P<0.05).There was no significant difference between the anti-inflammatory effect of Natural Group,Biological Group and Chemical Group (P>0.05).Conclusion Resina Draconis generated by different artificial induction methods proves itself with no significant difference in anti-inflammatory and analgesic effects.
