Abstract:Objective To research the effect of lung cancer mdig gene (mineral dust-induced gene)on methylation status of lysine residues on histone H3 and H4 partial loci,and to provide basis for diagnosis and treatment of lung cancer.M ethods 200 patientswith lung cancer(T1-T4)treated in our hospital from December 2010 to December 2015 were randomly selected,and UTX,Rb mRNA and protein content of cancer tissue and para-carcinoma tissue were detected,as well as the H3K27me3 and Rb protein content,and activity of UTX demethylase of cancer tissue and paracarcinoma tissue,to determine the effect of UTX expression and dysfunction in development,progress,and outcome of lung cancer.Results The UTX and Rb ofmRNA and protein content of cancer tissue and para-carcinoma tissue were various among patientswith lung cancer at different stages,and therewere significant differences in H3K27me3 and RB protein content,and activity of UTX demethylase of cancer tissue and para-carcinoma tissue in lung cancer patients(P<0.05).According to the result of H3K27me3 content detection in lung cancermdig gene lung carcinoma cells,there was no significant difference in H3K27me3 content between silentmdig lung carcinoma cells and over-expressedmdig lung carcinoma cells (P>0.05).Conclusion Histone demethylase,UTX,and Rb play an important role in development and progress of lung cancer tissues by inhibiting development of cancer cells and controlling spread of cancer cells.More studies on methylation status of lysine residues on histone H3 and H4 partial loci have significantmeanings for treatment and diagnosis of lung cancer.
