Application value of fluorescence PCR melting curve analysis in detecting tuberculosis drug resistance

Abstract:Objective To investigate the detection efficiency of Rifampicin and Isoniazid related drug resistance genes in sputum samples by fluorescence PCR melting curve analysis.Methods The sputum test results of 254 cases in the 10th People′s Hospital of Shenyang City from July 2019 to July 2020 were retrospectively analyzed.Using BACTEC MGIT 960 liquid drug sensitivity (hereinafter referred to as MGIT 960 drug sensitivity) as the reference standard, the effectiveness of the fusion curve method in the detection of Rifampicin and Isoniazid resistance were evaluated.Results Compared with MGIT 960, the sensitivity, specificity and Kappa values of Rifampicin resistance were 89.6%, 95.6%and 0.83.The sensitivity, specificity and Kappa values of Isoniazid resistance were 81.5%, 98.9% and 0.85.In the initial treatment of patients with good consistency, better than the retreatment of patients.Conclusion The method to detect patients with sputum samples of Rifampicin and Isoniazid resistance genes, compared with MGIT 960 DST method has good sensitivity and consistency, specific degree, can quickly detect patients with drug resistance situation and multidrug-resistant tuberculosis (MDR-TB) patients, especially for clinical patients to treat patients with early reference provides a quick and effective chemotherapy scheme.
