To explore the PCR-RFLP in testing the relationship between methylenetetrahydrofolate reductase gene polymorphism and the pathogenesis of pre eclampsia |

Abstract ObjectiveTo investigate the restriction fragment length polymorphism polymerase chain reaction(cleaved amplification polymorphism sequence-tagged sites,PCR-RFLP)in testing the relationship between methylenetetrahydrofolate reductase gene polymorphism and the pathogenesis of pre eclampsia,provide the basis for the prevention and treatment of preeclampsia.Methods100 preeclampsia (the case group)and 500 healthy pregnant women (the control group)from May 2016 to June 2017 treated in our hospital were selected as subjects to conduct polymorphism analysis of methylenetetrahydrofolate reductase gene and detect of gene polymorphism.Blood biochemical indexes were analyzed,basic data was investigated and factors were analyzed.ResultsAll cases were successfully sequenced in the case group,and 498 cases were successfully sequenced in the control group.The genotype frequency and allele frequency of the case group were compared,and the differences were statistically significant(P<0.05).The the number of junior high school degree and below level,planned pregnancy,planned to control diet and nutritional supplements,folic acid,vitamin D supplements,the proportion of malnutrition were compared,and the differences were statistically significant(P<0.05).APPT in the case group was lower than that in control group,and Hcy was higher than that in control group,the differences were statistically significant(P<0.05).Logistic regression analysis was used for multivariate analysis.The results showed that allele TT,folic acid supplementation and planned exercise during pregnancy were independent risk factors(P<0.05).There was significant correlation between TT type and Hcy and APPT(P<0.05).ConclusionMethylenetetrahydrofolate reductase gene polymorphisms are associated with preeclampsia and have independent effects,which may be related to the elevation of Hcy.
