Influence of Caffeic Acid on blood glucose concentration in rats with type 2 diabetes mellitus

Abstract:Objective To explore the influence of Caffeic Acid on blood glucose concentration in rats with type 2 diabetes mellitus.Methods Sixty male SD rats aged 6-8 weeks were selected and fed for 3 days.Twenty rats were selected as the normal observation group and they were divided into the blank group and experimental group1-3,and there were 5 rats in each group.The experimental group 1~3 was fed with 0.4,0.6 and 0.8 g/kg of Caffeic Acid respectively,the blank group was gavaged with 1 ml/100 g of equivalent distilled water.The other 40 rats were fed with high-sugar and high-fat diet for 6 weeks and intraperitoneally injected with 30 mg/kg Streptozotocin(STZ)for 3 days,the model of type 2 diabetic rats was established,FPG>7 mmol/L and rat urine sugar test positive were considered to be successful in modeling.Thirty rats successfully modeled were randomly divided into model group,Acarbose group,combined group and Caffeic Acid group 1-3,and there were 5 rats in each group.Acarbose group was given 10 mg/kg Acarbose,combined group was given 10 mg/kg of Acarbose and 1 g/kg of Caffeic Acid,Caffeic Acid group 1-3 was given 0.8,1.0,1.2 g/kg respectively,model group was given 1 ml/100 g of distilled water.The influence of Caffeic Acid on fasting blood glucose(FPG)and insulin(INS)in rats of normal observation group was observed at different times;the influence of each drug group on the concentration of FPG in type 2 diabetic rats was observed.Results There was no significant difference in FPG concentration and INS expression level among the groups (in normal observation group)before gavage,gavage for the first,sixth day(P>0.05).There was no significant difference in FPG concentration among the groups(in model group and various drug group)before gavage(P>0.05).After gavage,compared with model group,the FPG concentration in various drug group were decreased significantly,and the differences were statistically significant(P<0.05,P<0.01).The FPG concentration in combined group was significantly lower,and the difference was statistically significant(P<0.01).Among the drug groups,the combined group had the most significant hypoglycemic effect(P<0.05).Conclusion Caffeic Acid has a reducing FPG effect on type 2 diabetic rats,but it is not as effective as Acarbose in reducing blood sugar.The effect of Acarbose combined with Caffeic Acid was the most obvious.
