Effect of MTH1 inhibitor TH588 on the proliferation and migration of human malignant melanoma A375 cells |

Abstract Objective To investigate the effect of MTH1 inhibitor TH588 on the proliferation and migration of human malignant melanoma A375 cells.Methods Cell culture method was used to culture human malignant melanoma A375 cells,the inverted microscope method was used to observe the influence of TH588 on cell morphology,and the CCK-8 method was used to detect the inhibitory effect of different concentrations of TH588(0,4,8,16,32,64 μmol/L)on the proliferation of A375 cells.The colony cloning inhibition test was used to observe and compare the proliferation inhibition effect of TH588 on A375 cells at three concentrations(12.8,6.4 and 3.2 μmol/L),cell scratch experiment was used to detect the effect of TH588 on the migration ability of A375 cells.Results The CCK-8 test results showed that the cell survival rate of the TH588 groups with different concentrations at the same time period was lower than that of the blank control group.The cell survival rate at the concentration of 32 and 64 μmol/L TH588 at 48 and 72 h was lower than that at 24 h.The results of the inverted microscope method showed that after TH588 acted on human malignant melanoma A375 cells,the cells shrank significantly,the edges became blurred,and debris particles were produced.Colony cloning inhibition test showed that TH588 had a significant inhibitory effect on the proliferation of human malignant melanoma A375 cells.The cell scratch experiment showed that the scratch migration distance of A375 cells after TH588 was shorter than that of A375 cells without intervention within 48 h.Conclusion TH588 has a significant inhibitory effect on the proliferation and migration of human malignant melanoma A375 cells,and low concentration TH588 has a better inhibitory effect.
