The effect of cryoprecipitation dissolution with dry blood soluble machine and water-bath blood soluble machine and the length of dissolution time on the biological activity of coagulation factor in cryoprecipitate

Abstract:Objective To investigate the use of dry and water-bath blood soluble machine for cryoprecipitation dissolution and the effect of length of dissolution time on the biological activity of coagulation factors in cryoprecipitate,in order to provide the basis for better results.Methods Altogether 120 bags of cryoprecipitate (Specification:1.0 U/Bag) with similar blood collection dates (less than 5 days) that were provided by our Central Blood Station of Baoan from February 3rd to February 7th in 2016 were selected and randomly divided into two groups (group 1 and group 2),with 60 bags in each group.group 1 adopted dry blood soluble machine for cryoprecipitation dissolution;while,group 2 adopted water-bath blood soluble machine for cryoprecipitation dissolution.FactorⅧ (FⅧ),von Willebrand factor antigen (vWF-Ag),prothrombin time (PT),thrombin time (TT),activated partial thromboplastin time (APTT) and plasma fibrinogen (Fib) of 120 cryoprecipitate samples that were placed in different blood soluble machines at 37℃ for 10,20 min and 30 min were determined.Results There was no statistically significant difference between group 1 and group 2 in the effects of blood plasma dissolution and all the indexes (P>0.05);however,with the prolonging of dissolution time,prolonged APTT as well as decreased activity of FⅧ and VWF antigen were seen,with statistically significant difference (P<0.05).Conclusion Prolonged blood plasma dissolution time will lead to decreased activity of FⅧ and VWF antigen as well as prolonged APTT,with statistically significant difference.To achieve better efficacy of clinical blood transfusion,the best time for cryoprecipitation dissolution is recommended to be 10 min.
