Abstract:Objective To establish an ultra performance liquid chromatography-tandem mass spectrometry method for the determination of four phenolic acid components (chlorogenic acid, ferulic acid, protocatechuic acid and protocatechualdehyde) in Cassia obtusifolia.Methods Cassia obtusifolia was extracted by ultrasound with 50%methanol.Chromatographic separation was performed on Acclaim 120 C18 (150 mm×4.6 mm, 5 μm) column by gradient elution with the mobile phase consisted of 0.1% formic acid (A) and acetonitrile (B).The flow rate was 0.5 ml/min, the column temperature was 30℃, and the injection volume was 2 μl.The mass spectra was obtained by electron spray ionization (ESI) under negative ion mode.Results The linear ranges of chlorogenic acid, ferulic acid, protocatechuic acid and protocatechualdehyde fell into 0.0200-3.2000,0.0230-3.6800,0.0230-3.6800,0.0280-4.4800 μg/ml,respectively.The average recovery of chlorogenic acid,ferulic acid,protocatechuic acid and protocatechualdehyde was 96.8%, 99.2%,96.3%and 96.9%,respectively.The contents of chlorogenic acid,ferulic acid,protocatechuic acid and protocatechualdehyde were 85.691-304.280, 5048.3-139 69.0, 1458.2-9919.4 and 15.108-22.956 μg/g, respectively.Conclusion This method can be used for the determination of four phenolic acids in Cassia obtusifolia, which lays a foundation for the study on the dose-effect relationship of antioxidant activity of Cassia obtusifolia.
