Exploration of open field test evaluating the mental fatigue of rats with spleen qi and spleen yang deficiency

Abstract:ObjectiveTo explore the possibility and the preliminary mechanism of open field test evaluating the mental fatigue of rats with spleen qi deficiency(SQD)and spleen yang deficiency(SYD).Methods24 SPF male SD rats were selected as objects and they randomly divided into control group,SQD and SYD group,each group had 8 rats.The OFT method was used to detect the motion distance and vertical times.Electron microscope was used to observe the mitochondrial morphology of hippocampal neurons.ATP level in hippocampal tissues of each group were measured by colorimetry.ResultsMotion distance in open field test of SQD and SYD group was shorter than that of control group,and vertical times in SQD and SYD group was fewer than that of control group,and there was a statistical difference(P<0.05).Motion distance in open field test of SYD group was shorter than that of SQD group,and vertical times in SYD group was fewer than that of SQD group,and and there was a statistical difference(P<0.01).Hippocampus neuron mitochondria in SQD and SYD group showed swelling and vacuolization,and ATP level in SQD and SYD group were significantly lesser than that in control group(P<0.01),and ATP level in SYD group was lower than that in SQD group(P<0.01).ConclusionOFT can be used to evaluate and rank the mental fatigue of rats of SQD,SYD objectively and its biological bases might related to the damage of hippocampus neuron mitochondria.
