紫草油对肛瘘术后组织修复及TGF-β1和Smad3表达的影响

Abstract Objective To explore the mechanism of Arnebia Root Oil promoting wound healing by studying the expression changes of TGF-β1and Smad3 factors after anal fistula surgery.Methods A total of 90 anal fistula patients admitted to our hospital from March 2017 to March 2018 were enrolled in this study.They were divided into the Arnebia Root Oil group(n=30),Fuzhiqing group(n=30)and Vaseline group(n=30)according to the simple random method.In the Arnebia Root Oil group,Arnebia Root Oil was used,while in the Fuzhiqing group,Fuzhiqing for external application was used,and in the Vaseline group,Vaseline for external application was adopted.Expression of TGF-β1and Smad3 factors among three groups was compared and detected by reverse transcription-polymerase chain reaction(RTPCR)and Western blot.Results RT-PCR and Western blot showed that the content of TGF-β1in the Arnebia Root Oil group increased gradually.On the 21stday after treatment,the content of TGF-β1in the Arnebia Root Oil group was higher than that in the Fuzhiqing and Vaseline groups with statistical significances(P<0.05).On the 21stday after treatment,the content of Smad3 in the Arnebia Root Oil group was lower than that in the Fuzhiqing and Vaseline groups,which differed statistically (P<0.05).Meanwhile,the healing time in the Arnebia Root Oil group was shorter compared with that of the other two groups (P<0.05).Conclusion Arnebia Root Oil can effectively promote wound healing after anal fistula surgery,which may be regulated by TGF-β1up-regulation and Smad3 down-regulation,thus providing a preliminary theoretical basis for Arnebia Root Oil treatment after anal fistula surgery identified as a damp-heat pouring downward pattern.
