肿瘤相关巨噬细胞表达miRNA-122对肝癌患者的作用

Abstract Objective To analyze the effect of miRNA-122 expression in tumor associated macrophages(TAM)on hepatocellular carcinoma patients.Methods A total of 20 cases of patients with hepatocellular carcinoma dianosed in our hospital from January 2015 to January 2018 were selected as the HCC-1 group,and another 20 healthy people were selected as the C-1 group.The peripheral blood monocytes in the two groups were collected and the level of serum miRNA-122 was tested by q-PCR technology.Forty healthy SD nude mice were selected and randomly divided into the HCC group(n＝20)and Scamble group(n＝20).The HCC group was injected subcutaneously with 0.5 ml Hep-2 cell suspension of liver cancer,while the Scamble group was given 0.5 ml normal saline.After modeling,arterial blood was collected from two groups of nude mice by abdominal femoral artery blood sampling for 5 consecutive days,and peripheral mononuclear cells were isolated and obtained.The level of miRNA-122 in TMA was tested by q-PCR quantitative analysis technique.Results The level of miRNA-122 in the HCC-1group was significantly lower than that in the C-1group,and the difference was statistically significant(P＜0.05).Two days after modeling,the level of miRNA-122 in TAM of nude mice in the HCC group was significantly lower than that in the Scamble group,and the difference was statistically significant(P＜0.05).Conclusion The level of miRNA-122 in TAM of liver cancer patients and liver cancer nude mice is lower.The miRNA-122 plays a role in regulating the growth of liver tumors.
