Study on boiling and dispersing technology of Tongbi Huoluo Decoction |

Abstract Objective To explore the decoction technology of Tongbi Huoluo Decoction.Methods The content of osthole and paeoniflorin in Tongbi Huoluo Decoction were determined by high performance liquid chromatography (HPLC)with XBridge C18 column,acetonitrile-water (49:51) as mobile phase,detection wavelength at 330,230 nm and column temperature at room temperature of 25℃.The contents of total solids and index components were used as evaluation indexes,and orthogonal test was used to screen the decoction process.By comparing the quality of decoction with that of traditional decoction pieces,the dosage of decoction was screened.Results Ostholewas in the range of 0.1224-4.0800 μg,with a good linear relationship (r=0.9999),the average recovery rate was 101.14%,RSD was 1.42%.Paeoniflorin has a good linear relationship in the range of 0.8194-16.3880 μg (r=0.9999),with an average recovery of 99.55% and RSD of 1.79%.The best decocting process was to add 12 times of water,soak for 5 min,decocting for 20 min each time,and decocting once.Through screening the clinical recommended dose test of decoction,the decoction dose equivalent to 35% of the traditional decoction pieces of Tongbi Huoluo Decoction was selected as the clinical recommended dose.Conclusion The optimized decocting process is simple,rapid,stable and feasible,and the selected dosage can provide guidance for clinical application.
