Abstract ObjectiveTo investigate the hypoglycemic effects and the function of islet β cells of Alogliptin combined with Glimepiride in type 2 diabetes mellitus rats.Methodsfifty rats of type 2 diabetes mellitus which were induced through high-sucrose and high-fat diet combined with low-dose abdominal injection of Streptozotocin were randomly divided into five groups:model group(group A),Alogliptin group(group B),Glimepiride group(group C),Alogliptin combined with Glimepiride group (group D),Alogliptin combined with Glibenclamide (group E),10 rats in each group.Another 10 normal rats were selected as normal group(group F).The fasting blood glucose(FBG),level of plasma insulin(FIns),total cholesterol(TC)and triglyceride(TG)levels were detected in rats after treated for 28 days,the HOMA-βF index and insulin resistance index(HOMA-IR)were calculated.ResultsAfter 28 d treatment,the FBG,TG,TC,Fins and HOMAIR of the rats in group A,B,C,D and E were all higher than those in group F,the HOMA-βF was lower than group F,and the differences were statistically significant(P<0.01).The FBG,TG,TC,FIns,and HOMA-IR of rats in group B,C,D and E were significantly lower than those in group A,and HOMA-βF was higher than group A,and the differences were statistically significant(P<0.01).The FBG and FIns of rats in group C were lower than those in group B,TG and TC were higher than those in group B,the differences were statistically significant(P<0.05).The FBG,TG,TC,Fins and HOMA-IR of rats in group D and E were significantly lower than those of group B,HOMA-βF was higher than group B,and the differences were statistically significant(P<0.01).The FBG,TG,TC,FIns and HOMA-IR of the rats in group D and E were lower than those in group C,HOMA-βF was higher than that in group C,and the differences were statistically significant(P<0.05).There was no statistically significant difference between D and E groups(P>0.05).ConclusionAlogliptin,Glimepiride single or combined can significantly reduce the blood glucose of type 2 diabetic rats,improve TG,TC,the function of islet β cell and insulin resistance,but combined effect is better than single drug therapy.
