UPLC method in the determination of the content of Vitamin E in plasma |

Abstract ObjectiveTo establish a rapid and accurate method in the determination of the content of Vitamin E in children′s plasma clinically.Methods200 μl children′s plasma was collected.140 μl methanol and isopropanol were added with the proportion of v/v,8:2,which were vortexed for 30 s.1 ml n-hexane was added and vortexed for 1 min.They were centrifuged at a high speed at room temperature for 120 s(12 000×g).700 μl supernatant was collected after centrifuge,blown dry and redissolved by 100 μl methanol.Chromatographic condition:BEH C18chromatographic column from Waters company was applied (50 mm×2.1 mm,1.7 μm).The prepared solution of 5 μl was taken as a sample,and determined with the flow rate of 0.3 mL/min.Methanol-water(96:4)was the mobile phase.The column temperature was 30℃.Under the condition,the determination wavelength was 291 nm.ResultsThe chromatographic peak of vitamin E in plasma was separated favorably,and non-interference linear regression equation was y=118296x+11644,r2=0.9997.The recovery rate was 90%-120%.ConclusionThe method is simple to operate,which is suitable for the rapid determination of Vitamin E content in children′s plasma.
