Effect of electroacupuncture on urodynamics and mechanism of neurogenic bladder retention after sacral pulp injury in rats |

Abstract Objective To study the effect and mechanism of electroacupuncture on urodynamics of neurogenic bladder retention after sacral pulp injury in rats. Methods Thirty healthy male rats were selected and divided into model group,control group and electroacupuncture group according to random number table method, with 10 rats in each group. The control group was not modeled or treated, and the model group was only modeled and not treated, and the electroacupuncture group received electroacupuncture treatment after modeling. After treatment, the urodynamic changes and other related index in the three groups of rats were detected. Results Compliance, leakage point pressure, basic bladder pressure and maximum bladder volume in model group and electroacupuncture group were higher than those in control group, and the differences were statistically significant (P<0.05). The above indexes in electroacupuncture group were lower than those in model group, and the differences were statistically significant (P<0.05). The mRNA levels of heat shock protein 20 (HSP-20), phosphatase and tensin homologous protein (PTEN) in spinal cord tissue of rats in model group and electroacupuncture group were higher than those in control group, and the differences were statistically significant (P<0.05). The mRNA level of HSP20 in electroacupuncture group was higher than that in model group, while the mRNA level of PTEN was lower than that in model group, with statistical significance (P<0.05). The levels of Blymphocytoma-2 gene (Bcl-2) and protein kinase B (Akt) in model group and electroacupuncture group were lower than those in control group, the differences were statistically significant (P<0.05). BCL2-associated X protein (Bax),FOXO1 and caspa se -9 in model group and electroacupuncture group were higher than those in control group, the differences were statistically significant (P<0.05). The levels of Bax, FOXO1 and Caspase-9 in electroacupuncture group were lower than those in model group, while the levels of Bcl-2 were higher than those in model group, the differences were statistically significant (P<0.05). There was no significant difference in Akt between electroacupuncture group and model group(P>0.05).Conclusion Electroacupuncture treatment of neurogenic bladder urinary retention rats after sacral spinal cord injury can improve their urodynamics. The specific mechanism of action can be analyzed and inhibited PTEN expression, promoted HSP20 phosphorylation and expression, and further inhibited apoptotic pathways and increased Akt activity Relevant.
