The role of cell apoptosis induced by endoplasmic reticulum stress in mice with deep vein thrombosis |

Abstract Objective To explore the role of cell apoptosis induced by endoplasmic reticulum stress(ERS)in mice with deep vein thrombosis(DVT).Methods Thirty-two mice were randomly divided into the normal group(10 mice),the sham operated group(11 mice)and the model group(11 mice).The model group used the method of inferior vena cava stenosis to construct the DVT model of mice.The sham operated group only exposed the vein through laparotomy without ligation.Fluorescence intensity of glucose regulated protein 78(GRP78)was detected by immunofluorescence technique.The expression of GRP78,X-box binding protein 1(XBP1),CCAAT-enhancer-binding protein homologous protein(CHOP)mRNA was detected by Realtime PCR.The expression p53 up-regulated modulator of apoptosis(PUMA),cytochrome C(Cyt C)and cysteinyl aspartate specific proteinase-3(cleaved caspase-3)was detected by western blot.Results Fluorescence intensity of GRP78 and GRP78,XBP1,CHOP mRNA expression volume of the model group were higher than those of the normal group and the sham operated group,the differences were statistically significant(P<0.05).PUMA,Cyt C and cleaved-caspase 3 protein expression levels of the model group were higher than those of the normal group and the sham operated group,the differences were statistically significant(P<0.05).Conclusion These results suggested ERS might be an important cause of apoptosis in DVT mice.
