Effect of TNIP1 gene silence on the expression of BCR and CD40 gene in B cells of patients with systemic lupus erythematosus |

Abstract Objective To explore the effect of TNIP1 gene silence on the expression of BCR and CD40 gene in B cells of patients with systemic lupus erythematosus (SLE).Methods Blood samples from 20 SLE patients and 20 healthy subjects received in Wuhan University Hospital during March 2018 were collected.Lentiviral-mediated TNIP1 gene silence was used as an entry point.Real-time quantitative PCR was used to detect mRNA expression level as well as expression levels of BCR and CD40 in TNIP1 gene in cells.Results The expression level of TNIP1 gene was higher in SLE patients, and interference with TNIP1 gene in B cells could lead to down-regulation of BCR and CD40 expression in B cells.Conclusion The expression of TNIP1, BCR and CD40 genes in B cells show a consistent trend, suggesting that TNIP1, BCR and CD40 have a certain regulatory relationship.
