Intervention effect of β-elemene on the expression of VEGF in the retina of rats with diabetic retinopathy during proliferating phase |

Abstract ObjectiveTo observe the effect of β-elemene on the expression of vascular endothelial growth factor(VEGF)in the retina of rats with proliferative diabetic retinopathy caused by streptozocin(STZ),to investigate its possible mechanism of action on retinopathy of rats with proliferative diabetic retinopathy.MethodsA total of 80 Wistar rats were selected.They were randomly divided into normal control group (NC group),STZ-induced diabetes model group(DC group),low-dose β-elemene group(BL group)and high-dose group(BH group),20 rats in each group.In the administration group,different doses of β-elemene and hematoxylin-eosin staining were injected into the vitreous cavity under the surgical microscope respectively.The pathological changes of rat retina tissues were observed.Retinal slices from each experiment group were collected.Immunohistochemical staining was applied to VEGF,and the expression of VEGF in retinal tissues was detected.ResultsThe results of HE staining showed that the tissue structure of the retina in the NC group showed a clear level.The retina was thinner in DC group than that in the NC group,and the structure was broken.The number of ganglion cells was reduced;the BL group had a slight increase compared with the DC group.The retinal structure in the BH group was better than that in the BL group.After immunohistochemical staining,the retinal results in each experimental group showed that the number of VEGF-positive nuclei in the DC group,NC group,BL group and BH group was (125.4671±8.4561),(10.1324±2.0334),(75.5123±7.6507),(21.5364±1.7861)respectively,differences between the groups were statistically significant(P<0.05).Conclusionβ-elemene may become a new target for the treatment of proliferative diabetic retinopathy.At the same time,it can improve the development of advanced DR and delay the further deterioration of retinal tissue.
