Abstract:Objective To explore the cytotoxicity of fibrin glue to human adipose stem cells by CCK-8 method. Methods Human adipose stem cells were obtained from the waste adipose tissue of a 28-year-old healthy woman who came to our hospital on November 12, 2018 for adult liposuction, and made into the 50% fibrin glue leaching solution group,the 100% fibrin glue leaching solution group, the negative control group and the positive control group respectively.The absorbance values in each fibrin glue medium was determined by CCK-8 method.Results The growth of primary adipose stem cells (ASCs)was slowly.The growth of ASCs was accelerated after passage, and ASCs appeared fibroblastlike morphology.There were no significant differences in the absorbance between the 50% fibrin glue extract group,the 100%fibrin glue extract group and the negative control group at 24 and 72 hours (P>0.05).The absorbance values of 50%and 100% fibrin glue extract group and the negative control group were lower than that of the positive control group,with statistical significance (P<0.05).Conclusion Fibrin glue has no significant cytotoxicity when mixed with ASCs,which can provide a reference for the exploration of adipose structure based on fibrin glue as scaffold material.
