Impact of Valproic Acid on drug resistance of HL-60 cell

Abstract:ObjectiveTo explore the impact of Valproic Acid(VPA)on drug resistance of HL-60 cells(HL-60/HT)resistant harringtonine.MethodsHL-60/HT cell strain was established by incremental pressure culture and divided into 3 groups:Ara-C group,VPA group and VPA+Ara-C group.MTT method was used to determine the change of proliferation of sensitive cells and drug resistant cells after using drug.The change of VPA resistance in HL-60/HT cells was determinated simultaneously.The cell cycle of seneitive HL-60 cells and HL-60/HT cells were detected by flow cytometry.ResultsVPA in combination with Ara-C can obviously inhibit the growth of HL-60 and HL-60/HT cell,and the inhibition rate was higher than that of VPA or Ara-C alone(q=1.37,1.51).ConclusionVPA can inhibit the growth of HL-60 and HL-60/HT cell.It can play a synergistic role with Ara-C and reduce the drug resistance of leukemia cells.
