Abstract:Objective To study the effect of tumstatin 19 peptide proliferation and apoptosis of hepatoma SK-Hep-1 cells. Methods CCK-8 was used to detect the effect of 19 peptide (0, 50, 100, 150, 200, 250 μg/ml) on the proliferation of SK-Hep-1 cells. DAPI staining was used to detect the morphological changes of SK-Hep-1 cell apoptosis,and flow cytometry was used to detect the effect of 19 peptide on SK-Hep-1 cell apoptosis. Results Sk-hep-1 cells were treated with 0, 50, 100, 150, 200, 250 μg/ml 19 peptide, after 24 h, the survival rates were (100.00±1.80)%, (95.64±1.92)%, (82.18±1.67)%, (68.43±1.39)%, (46.74±1.35)%, (42.49±1.85)%, respectively. After 48 h treatment, the cell survival rate was (100.00±1.80)%, (87.27±1.96)%, (66.32±1.74)%, (58.78±1.94)%, (40.40±1.44)%, (22.15±1.51)%, respectively. When 19 peptide was 0 μg/ml, most of SK-HEP-1 cells were stained uniformly and blue, and apoptosis did not occur. Compared with 0 μg/ml, some cells showed bright blue after treated with 50, 100, 150, 200 and 250 μg/ml 19 peptide for 24 h, and the nuclei showed concentration, rupture, fragmentary dense and dense staining or cap-like apoptotic morphology. The apoptosis rate increased with the increase of 19 peptide concentration. Conclusion 19 peptide can effectively inhibit the proliferation of SK-Hep-1 cells and induce the apoptosis, which provides theoretical guidance for the research and clinical treatment of new anti hepatoma drugs.
