Abstract Objective To investigate the expression of BUB3 and Bub1 related proteins(BubR1)in the villus and decidual tissues of patients with recurrent miscarriage (RM)and their relationship with RM.Methods From March 2016 to March 2018,50 cases of missed abortion pregnant women in Huadu District People′s Hospital of Guangzhou were selected as the case group,and 50 cases of normal induced abortion women in the same period were selected as the control group.The age,body mass index(BMI),parity,total cholesterol(TC),triglyceride(TG),low density lipoprotein cholesterol(LDL-C),high density lipoprotein cholesterol (HDL-C) were collected.The mRNA and protein levels of BUB3 and BuBR1 in embryonic villus tissue(VT)and decidual tissue(DT)were determined by real-time quantitative fluorescence PCR(qRTPCR) and Western blot (WB) Logistic regression was used to analyze the influencing factors of RM.Results There were no significant differences in age,BMI,parity,TC,TG,LDL-C and HDL-C levels between the two groups(P>0.05).The mRNA and protein expression levels of BUB3 and BubR1 in VT and DT in case group were lower than those in control group,and parity was more than that in control group,and the differences were statistically significant (P<0.05).Logistic regression showed that the parity (β=0.461,OR=1.586,95%CI=1.253-2.007),BUB3 mRNA (β=0.710,OR=2.035,95%CI=1.602-2.585) and BubR1 mRNA (β=1.304,OR=3.685,95%CI=2.884-4.708) were independent risk factors for RM (P<0.05).Conclusion The mRNA and protein levels of BUB3 and BubR1 are reduced in VT and DT of patients with RM.BUB3 and BubR1 mRNA are independent risk factors for the occurrence of RM and may be potential diagnostic indicators for RM.
