Study of Curcumin on hepatic stellate cells in rats |

Abstract ObjectiveTo explore the mechanism of Curcumin on the prevention and treatment of liver fibrosis in rats,so as to provide reference for clinical treatment.MethodsThe rat model of experimental liver fibrosis induced by CCl4was established.At the same time,20 mg,10 mg and 5 mg Curcumin were in lavage in every 100 g body weight,the corresponding models were set as delivery model group,model group and control group respectively,10 rats in each group.The level of reactive oxygen species(ROS)in serum was detected by colorimetric method.Malondialdehyde(MDA)content in liver tissue was detected by MDA assay kit(TBA method).By electrophoretic mobility shift assay(EMSA),the activity of NF-κB in liver tissue was detected.The levels of NF-κB expression,TGF-β1,platelet-derived growth factor-BB(PDGF-BB)and recombinant human connective tissue growth factor(CTGF)in liver tissue were detected by immunohistochemistry.The mRNA level of tissue inhibitor of metalloproteinase-1 in liver tissue was detected by RT-PCR.The results after detection were statistically analyzed.ResultsIn the model group,the levels of ROS,endotoxin,MDA,NF-κB,TGF-β1,PDGF-BB,CTGF and TIMP-1mRNA were significantly higher than those in the normal group,and the differences were statistically significant(P<0.05).The levels of above-mentioned factors in the delivery model group were all lower than those in the model group,and the differences were statistically significant(P<0.05).ConclusionCurcumin presents with the inhibition of ROS and endotoxin levels in serum,MDA,NF-κB,TGF-β1,PDGF-BB,CTGF,and TIMP-1 mRNA expression in liver tissue to some extent and participants in the liver fibrosis prevention through a variety of mechanisms.
