Regulation of Oxytocin on feeding in the amygdaloid nucleus |

Abstract Objective To observe the regulation of Oxytocin (OT)on feeding in the amygdaloid nucleus.Methods Forty-eight male Sprague-Dawley(SD)rats were selected.Among them,16 rats were randomly divided into free-feeding group and 72-hour fasted group,with 8 in each group.Immunohistochemistry was used to detect the expression of OT receptors in the rat amygdala.The remaining 32 rats were subjected to cannula implantation in amygdala and 16 of them were randomly divided into two groups,namely the injected with OT in amygdaloid nucleus group and the injected with normal saline in amygdaloid group 1 respectively,8 rats in each group.Food intake was measured in rats at 0.5,1,2,and 3 h.Another 16 postoperative rats were randomly divided into two groups,namely the injected with OT receptor blocker(Atosiban)in amygdaloid nucleus group and the injected with normal saline in amygdaloid group 2 respectively,8 rats in each group.Similarly,food intake of the rats was measured at 0.5,1,2,and 3 h.Results The expression of OT receptors in the amygdaloid nucleus of rats in the 72-hour fasted group was significantly higher than that in the normal feeding group,and the difference was statistically significant(P<0.05).The food intake of rats of injected with OT in amygdaloid nucleus group at 0.5,1,2,and 3 h were lower than those in the injected with normal saline in amygdaloid group 1,and the differences were statistically significant(P<0.05).The food intake of rats of injected with Atosiban in amygdaloid nucleus group at 0.5,1,2,and 3 h were higher than those in the injected with normal saline in amygdaloid group 2,and the differences were statistically significant(P<0.05).Conclusion OT participates in feeding regulation in the amygdaloid nucleus through acting on its receptors.
