甘油-卵黄-柠檬酸盐冷冻保护剂中添加二十二碳六烯酸对人精子冷冻前后运动学参数的影响

Abstract Objective To study the influence of addition of different molar concentration of docosahexaenoic acid(DHA)into Glycerol-Egg Yolk-Citrate(GEYC)cryopreservative mediums on the kinematic parameters of human sperm before and after cryosurgery so as to explore the application value of n-3 polyunsaturated fatty acids in cryosurgery of human sperm.Methods Altogether 33 cases of semen samples which came from semen donors during human sperm bank screening period in Guangzhou Family Planning Specialty Hospital from March 2016 to April 2017 were selected.Based on GEYC cryopreservative mediums,the concentration of exogenous DHA in mixed semen-GEYC liquor were 0 mmol/L in the control group,0.317 mmol/L in the DHA 1 group,0.634 mmol/L in the DHA 2 group and 1.268 mmol/L in the DHA 3 group.SCA 5.2 computer-assisted sperm analysis system was used to detect the sperm kinematics parameters 0-8 hours after addition of cryopreservation and frozen resuscitation.Results After addition of cryopreservation and frozen resuscitation,only curve line velocity(VCL)and average path velocity(VAP)of the DHA 3 group were significantly lower than those of the control group (P＜0.05),there was no statistically significant difference in kinematic parameters between the other groups(P＞0.05).After frozen recovery at 0,4,5,6,8 hours,only kinematic parameters of the DHA 3 group decreased occasionally,with statistically significant difference in contrast with 3 groups.After cryosurgery at 0-8 hours,most of the kinematic parameters had no statistically significant difference between each group(P＞0.05).Conclusion Addition of DHA into GEYC cryo-preservative mediums do not increase the sperm kinematic parameters before and after cryosurgery.The proper addition of DHA may affect frozen semen through other ways.Addition of a large number of DHA may have a negative impact on the part of the kinematic parameters before and after cryosurgery.
