Abstract Objective To discuss the effect of mesenchymal stem cell exosomes (MSCs-EXO)to neurological function recovery of rats with ischemic stroke.Methods Rat bone marrow mesenchymal stem cells were cultured in primary culture,MSCs-EXO were extracted by hyperspeed centrifuge method,and the rat ischemic stroke model was made by middle cerebral artery occlusion(MCAO)method,the MSCs-EXO were administered nasally and were set as MSCs-EXO treated group,and the neural function recovery was evaluated by modified neurological severity scores(mNSS)and misstep test,and compared with the untreated model group (MCAO group).The distribution of MSCs-EXO labeled by PKH26 in ischemic penumbra(IP) was observed by immunofluorescence staining.The expression level of PTEN in brain tissue in IP region was detected by fluorescence quantitative PCR and Western blot,and compared with normal rats and MCAO group.Results The recovery of nerve function in the MSCs-EXO treated group was higher than that in the MCAO group,and the difference was statistically significant (P<0.05).Immunofluorescence staining showed that labeled MSCs-EXO could enter nerve cells in the IP region.Quantitative fluorescent PCR and Western blot detection showed that PTEN levels in both the MSCs-EXO treated group and the MCAO group were higher than normal,but PTEN level in the MSCs-EXO treated group was lower than that in the MCAO group,with statistical significances(P<0.05).Conclusion MSCs-EXO can promote the recovery of neural function in ischemic stroke rats,which may be related to the down-regulation of PTEN expression in IP region.
